Sigma Collagenase Blends

In Sigma′s collagenase blends, collagenase and casein-digesting protease are separately prepared. They are then blended to give different collagenase:caseinase ratios. Collagenase activity is measure by the spectrophotometric FALGPA (N-[3-(-Furyll)acryloyl)]-Leu-Gly-Pro-Ala) assay, and caseinase activity by tyrosine release measured spectrophotometrically with the Folin-Ciocalteau reagent. Type F has the highest collagenase activity and lowest caseinase. Type L has high caseinase and low collagenase actitivities. Type H ratio has more nearly equal activities. Because optimizing the preparation of cells for culture varies greatly between tissue types, these blends are recommended for greater reproducibility between lots.
