Antibodies to ARTS:
A novel mitochondrial pro-apoptotic protein

Apoptosis-related protein in TGF-β signaling pathway (ARTS) is a novel mitochondrial protein that mediates apoptosis induced by a variety of pro-apoptotic stimuli. Originally found to be essential for apoptosis induced by transforming growth factor-β (TGF-β).1,2 ARTS was later shown to be a key apoptotic protein mediating apoptosis initiated by other inducers. The protein is derived form the human septin H5/PNUTL2/CSCrel-2b gene. It contains a phosphate-binding ({-loop) death domain, which is conserved in all septins and in many different classes of ATP/GRPases. Unlike other septins, ARTS is localized to mitochondria and translocates to the nucleus, coincident with apoptotic signals induced by TGF-β.2 In addition, mutation of GKS to ENP (amino acids 137-139) in the P-loop impairs the ability of ARTS to enter the nucleus and abolishes its ability to induce apoptosis.1,2 While the N-terminus of arts is homologous with hCDrel2b, the C-terminus contains a unique amino acid sequence that is not contained in either hrel2b or H5. ARTS induces apoptosis in response to TGF-β, while hCDCrel2b and H5 do not, suggesting the C-terminus of ARTS is necessary for its pro-apoptotic function.
