Presented by Dr. Fredrik Ponten, Uppsala University Hospital, Sweden The Human Protein Atlas project is based on a multi-disciplinary research program to allow for systematic exploration of the human proteome. An antibody-based proteomics strategy is applied to combine high-throughput generation of mono-specific antibodies (affinity-purified) with protein profiling in human tissues and cells using immunohistochemistry on cell and tissue microarrays (TMAs). Systematic exploration of the human proteome has provided a new insight into global protein expression patterns and identification of biomarkers with in silico based methods. To examine protein expression levels in tumors from large patient cohorts, special TMAs have been generated and immunostained with antibodies against selected targets. This allows for the analysis of protein expression levels with various clinico-pathological parameters including overall survival of respective patients. Using this strategy, new markers of potential clinical importance can be discovered and validated. Antibodies are also used for protein profiling of human plasma. A combination of undirected target selection, antibody suspension bead arrays and heat-induced epitope retrieval has allowed for a systematic analysis of protein signatures in plasma from patients representing major human diseases including various forms of cancer, cardio-vascular and neurodegenerative diseases. Dr. Ponten who is a former Fulbright scholar, is one of the founders of the Human Protein Atlas program and a member of the Atlas Antibodies AB Board and Human Proteome Resource Board. Dr. Ponten has been main supervisor for 10 PhD students (five finished) and is the author of 99 peer reviewed scientific papers and three book chapters. Dr. Ponten received his PhD from Uppsala University in 1996, he did a post-doctoral fellowship at Yale University (New Haven, CT, USA) and became full professor in 2008 at the Department of Genetics and Pathology where he now holds the chair. |
