Ethidium bromide is a potent mutagen, but is easily replaced by Nancy 520 - a safer chemical for nucleic acid staining. The Ames test results shown below illustrate how much safer we can make our staining procedures.

Nancy-520 fluorescent stain is specific for double-stranded DNA in agarose gels.

Higher sensitivity than ethidium bromide
  Ames II mutagenicity assay performed using Ethidium bromide (blue), SYBR Green I (red) and Nancy-520 (green). Nancy-250 produces fewer frame shift or base-pair substitution mutations than either ethidium bromide or SYBR Green I.

Excitation/Emission settings similar to SYBR Green I
   – no additional filters or light source needed
  DNA marker (Lambda DNA Hind III digest) at 200 ng/lane (left) and 20 ng/lane (right) was separated on a 1% agarose gel, stained with either Nancy-520 or SYBR Green I post-electrophoresis, and imaged. Nancy-520 produces sharp, clear bands with detection down to 0.84 ng of DNA.

   Linear detection range 0-2 µg/ml of DNA
  Quantitation of DNA (Lambda DNA Pst I Digest) in solution using Nancy-520, performed in a 96 well plate. Fluorescence was measured on a laser-scanner (FLA-3000, Fuji) with 532 nm excitation and 580 nm emission filter. Different concentrations of DNA in TE buffer, pH 7.5, and their corresponding fluorescence values are shown.
