Using the Beacon Designer 4 software from Premier BioSoft Int'l, our specialists provide you with an optimal design for your molecular beacons probe and primers pairs for real-time qPCR, offering you:
Optimal probe design: statistical optimization techniques are used to select probes free of dimers, repeats and runs to maximize signal strength.
Multiplex probes: We can design up to 4 sequences, which are non-homologous to prevent competition in multiplex reactions.
Allelic discrimination: We can design molecular beacons for detection of both wild type and mutant alleles.
Optimal probe Tm: by automatic selection of a stem of appropriate length.
Highly accurate hairpin Tm calculations are performed by connecting to the MFOLD server.
We can also evaluate your pre-existing molecular beacons to design the best primers to work with them.
Design Service Options
Sigma-Proligo™ can design AND synthesize your oligonucleotides, or just provide the design and leave the synthesis to you.
- Design AND synthesis service
Your oligonucleotides will be designed and synthesized (fee per oligonucleotide) within several working days of submitting your design request (3 days for design, plus 5 to 6 days for synthesis - pending successful QC validation). First design is FREE (excludes cost of synthesis).
- Design only service
Your design will be sent to you by email within 3 working days of submitting your design request. Complex primer and probe designs or multiplex assays can take longer to design.
Recommendations
We recommend using the following dye and quencher combinations:
- BHQ™-1 (3'end quencher) and 6-FAM™ (5'end reporter),
- TAMRA™ (3'end quencher) and 6-FAM (5'end reporter),
- DABCYL (3'end quencher) and 6-FAM (5'end reporter).
