Overview (ZFN)

Zinc finger nucleases (ZFNs) enable manipulation of the genome with unprecedented ease and precision. ZFNs are a class of engineered DNA-binding proteins that facilitate targeted editing of the genome by creating double-strand breaks (DSBs) in DNA at user-specified locations. DSBs are important for site-specific mutagenesis in that they stimulate the cell’s natural DNA-repair processes, namely homologous recombination and non-homologous end joining. Using well-established and robust protocols, these cellular processes can be harnessed to generate precisely targeted genomic edits resulting in cell lines, including somatic cell lines, with targeted gene deletions (knockouts), integrations, or modifications. Featured lectures will provide in depth discussion on the fundamentals of ZFNs as well as cutting edge applications for targeted gene knockout, integration and tagging. After mastering ZFN technology, registrants will have all of the tools they will need to modify genes in standard tumor cell lines, primary cells and embryonic stem cells. Registrants should be familiar with basic mammalian cell culture techniques.

Accommodations

The city of St. Louis offers a variety of convenient transportation and accommodation options. Lambert St. Louis International Airport (STL) is connected to downtown St. Louis by a major interstate (I-70) as well as a convenient Metrolink system that directly connects the airport terminal to multiple downtown stops and locations. Once in St. Louis, we strongly recommend staying at the right to Hilton at the Ballpark. Sigma has negotiated a special low rate for all of the RNAi workshop participants. In addition, a free shuttle will take all participants from the Hilton each morning to the Sigma Life Science and High Technology Center and then return them at the end of each day. This allows participants to focus on the RNAi course and not worry about scheduling transportation.
