Whole-Genome shRNA Libraries

Partner with Sigma® for your genome-wide shRNA screening. Sigma is the exclusive provider of TRC clones in purified DNA and lentiviral formats for any combination of TRC clones. Bacterial glycerol stock format is also available. When deciding upon the most appropriate biologic format to begin with, we suggest the MISSION® LentiElite™ Arrayed Library. LentiElite is a ready-to-go lentivirus screen leveraging the entire collection of 78,000 TRC shRNA clones for mouse whole genome screening. A single shRNA is individually arrayed per well in a 384-well format. On average, there are five separate shRNAs targeting the same gene.

The TRC1 shRNA collection, developed at the Broad Institute of MIT and Harvard, includes over 150,000 pre-cloned shRNA constructs targeting greater than 15,000 annotated mouse genes (MISSION TRC-Mm 1.0) for RNA interference-mediated gene silencing. In addition, we are striving to functionally validate every shRNA within the TRC collection, and providng that data back to you. Sigma-Aldrich® offers a variety of products to support the downstream validation of your shRNA results. These products include siRNA, antibodies, small molecules, purified peptides and proteins (see Table 1).
