The MISSION LentiPlex Pooled Library System Overview
The LentiPlex Pooled Libraries contain the entire TRC collection in 10 matrix tubes. The pooled virus is added to your cell population at a predetermined MOI. Cells are incubated then exposed to selective pressure and selected based on desired phenotype or flow sorting. After selection, cells are then isolated by colony cloning or FACS and the integrated shRNA is identified via PCR, microarray or "Next Generation" sequencing. The identity of potential "hits" can then be used to develop hypotheses regarding the biological role of the corresponding gene(s).

J774 murine macrophage-like cells were transduced with the pooled mouse shRNA library and analyzed by flow cytometry. Transduced cells were selected in puromycin prior to FACS analysis. Both transduced and untransduced cells were stained with a mixture of FITC- and APC-conjugated primary antibodies. Cells with strong APC and weaker FITC staining were discovered and isolated as "hits" in the screen. These cells are indicated by the arrow in the upper left side of the contour plot (right). The TRC-designed pLKO.1 vector of the LentiPlex pooled constructs enables the isolation of populations using most common fluorescent reporters including FITC and GFP.


Product Specifications
The MISSION LentiPlex Human and Mouse shRNA Pooled Libraries are genome-wide lentiviral pools produced using a proprietary process. The libraries each consist of over 75,000 shRNA constructs targeting 15,000+ human or 15,000+ mouse genes. Each library is tested for shRNA representation before product release to ensure robust library coverage. Each library is provided in ready-to-use lentiviral format at titers of at least 5 x 108 TU/ml via p24 assay and is pre-divided into 10 subpools of approximately 8,000 shRNA constructs each. Amplification and sequencing primers are also provided for downstream target identification.
