For Rapid, Convenient Genome-wide shRNA Screens
MISSION LentiPlex Pooled shRNA Libraries bring the power of the MISSION TRC shRNA collection together with Sigma's lentiviral manufacturing expertise to enable genome-wide RNAi screens on your bench-top. The LentiPlex pooled system provides enhanced delivery and long-term gene silencing in non-dividing and primary cell lines allowing unlimited discovery potential. The convenient format now makes rapid whole-genome RNAi screens accessible to any researcher with minimal reagent, time or capital equipment investment.
- Comprehensive – Trusted TRC content with shRNA targeting 20,000+ human or 21,000+ mouse genes
- Coverage – Average of 5–6 clones targeting each gene
- Quality – Proprietary methodology ensures uniformity and complete library coverage
- Universal – Simplified whole genome RNAi screening in standard or difficult to expand cell lines
- Accessible – Bench top discovery without robotics or liquid handling system
- Versatile – Compatible with FACS and most downstream fluorescent assays
The MISSION LentiPlex Human and Mouse Pooled shRNA consist of ~130,000 (over 43,000 validated) shRNA constructs targeting 20,000+ human genes and ~118,000 (over 36,000 validated) shRNA constructs targeting 21,000+ mouse genes. Each library is tested for shRNA representation before product release to ensure robust library coverage and is provided in ready-to-use lentiviral format at titers of at least 5 x 108 TU/ml via p24 assay. The MISSION LentiPlex libraries are pre-divided into various subpools (see Table 1 and Table 2 below for number of subpools and approximate number of shRNA constructs represented in each). Amplification and sequencing primers are also provided for downstream target identification.
- Pre-aliquoted into subpools ranging from 5,500 to 8,000 shRNA constructs per pool
- Enough material for six screens at 100X representation
- Titers of at least 5 x 108 TU/ml
- Amplification and sequencing primers provided

MISSION LentiPlex Pooled shRNA Human Library
Clone Coverage by Library
Table 1. The MISSION LentiPlex Human Pooled shRNA Library, Catalog Numbers SHPH01, SHPH15, SHPH2, SHPHLIBR are genome-wide lentiviral pools, powered by the RNAi Consortium (TRC). Each version of TRC (1.0, 1.5, and 2.0) corresponds to the LentiPlex Product Name and Number. The addition of LentiPlex 1.5 and 2.0 offer expanded gene and clone coverage from the original TRC1 library. The LentiPlex Complete consists of the full shRNA library of over 125,000 clones and targeting over 20,000 human genes.

The MISSION LentiPlex Pooled shRNA Library System Overview
The LentiPlex Pooled shRNA Libraries contain the TRC collection(s) in matrix tubes. The pooled virus is added to your cell population at a predetermined MOI. Cells are incubated then exposed to selective pressure and selected based on desired phenotype or flow sorting. After selection, cells are then isolated by colony cloning or FACS and the integrated shRNA is identified via PCR, microarray or "Next Generation" sequencing. The identity of potential "hits" can then be used to develop hypotheses regarding the biological role of the corresponding gene(s).
Figure 1. Flow Sorting of Cells Transduced with the MISSION LentiPlex Pooled shRNA Library
J774 murine macrophage-like cells were transduced with the pooled mouse shRNA library and analyzed by flow cytometry. Transduced cells were selected in puromycin prior to FACS analysis. Both transduced and untransduced cells were stained with a mixture of FITC- and APC-conjugated primary antibodies. Cells with strong APC and weaker FITC staining were discovered and isolated as "hits" in the screen. These cells are indicated by the arrow in the upper left side of the contour plot (right). The TRC-designed pLKO.1 vector of the LentiPlex pooled constructs enables the isolation of populations using most common fluorescent reporters including FITC and GFP.
