Quality Control:
- The amplified region of the cDNA with the highest possible number of highly effective siRNA is selected using DEQOR primer design program.
- PCR product of transcriptome library tested via gel electrophoresis and identity confirmed by DNA sequencing.
- Purified RNase III digested product (esiRNA) validated via gel electrophoresis.
- esiRNA concentration determined by measuring OD260 or fluorescence intensity.
Purification:
- RNase III digested products are purified via column purification with Q-Sepharose, isopropanol precipitation and ethanol wash.
Available MISSION esiRNA Products:
- 1 μg (~100 pmol) human genome scale esiRNA library: 1 μg of each esiRNA.
- 2.5 μg (~250 pmol) human genome scale esiRNA library: 2.5 μg of each esiRNA.
- 20 μg (~2 nmol) individual human and mouse esiRNA.
- 50 μg (~5 nmol) individual human and mouse esiRNA.
Format:
- All esiRNA are delivered in solution (resuspended in nuclease-free TE buffer) at the concentrations listed below, as measured by OD260 or fluorescence intensity.

- The standard plate format for MISSION esiRNA libraries is in 384-well plates, with all edge wells left empty. In addition, internal well positions D3,D4,D21,D22,M5,M6, M18, and M19 are empty. Individual MISSION esiRNA are provided in 96 well plates.
Shipping, Storage, and Stability:
- Shipped in solution on dry ice, in sealed plates.
- esiRNA stored at –20 °C have been demonstrated to be stable for 2 years. No decrease in stability after 12 freeze/thaw cycles.
Certificate of Analysis/Technical Data Sheet:
esiRNA will be delivered with an esiRNA Technical Data Sheet, which includes:
- Plate number and position
- esiRNA ID number
- Gene ID (Genbank) or Gene ID (Ensembl)
- Gene name
- Organism
- OD (units)
- Amount (μg)
- Concentration (ng/μL)
- Lot number
- Target sequence
- Additional data are provided for purified RNase III digested product (esiRNA) upon request.
- Custom esiRNA libraries and whole genome esiRNA libraries will be delivered with a CD containing complete plating information.
