The CompoZr Targeted Integration Kit utilizes a pair of Zinc Finger Nucleases (ZFN) targeted to the AAVS1 site on human chromosome 19. Using the AAVS1 ZFN you will introduce site-specific double strand breaks at the targeted locus, which with a co-transfected donor molecule, stimulates homology-directed recombination (HDR) and enables one to insert their gene of interest into this site for expression. Unlike spontaneous homologous recombination that occurs in mammalian cells at an extremely low rate, ZFN-mediated HDR achieves between 1%–20% targeted integration without selection in various cell lines, representing three to four orders of magnitude improvement in efficiency. With the CompoZr Targeted Integration Kit, you have the ability to insert your gene of interest into any human cell line without the requirement of engineering a transgene landing pad.
