S-Gal™/LB Agar Blends for Detecting Gene Insertions
|
|
|
S-Gal is a trademark of Sigma Chemical Co.
The evidence is clear: |
S-Gal produces the clearest contrast between lac+ and lac- colonies.

S-Gal™ a new chromogen to detect gene insertions:
|
- Contains a patented chromogenic substrate,
S-Gal (3,4-cyclohexenoesculetin-b-D-galacto- pyranoside).
- Replaces X-gal as the preferred substrate for
measurement of b-galactosidase transformed cells used in molecular biology applications.
- Hydrolyzed by b-galactosidase to yield an
intense black insoluble precipitate.
- Heat and light stable, non-toxic.
|
S-Gal, a new chromogenic substrate for gene insertion detection. |
S-Gal/LB Agar Blend
Product. No. C 4478 is a complete powder mixture useful for detecting prokaryotic cells which have been transformed with vectors containing the lacZ gene. It contains S-Gal, a new chromogenic substrate, and isopropyl b-D-thio-galactopyranoside, (IPTG).
IPTG initiates the transcription of the lacZ gene by binding to the lac repressor product of the Lac I gene and allowing the lac operator to turn on transcription. The product of the lacZ gene is b-galactosidase. b-Galactosidase transformed cells have traditionally been measured by the hydrolysis of X-gal (5-bromo-4-chloro-3-indolyl-b-D-galactoside) to a blue precipitate.
Now we have S-Gal!
| FEATURES |
BENEFITS |
| Faster colony color development. |
Pick colonies for faster results. |
| Lac+ colonies turn an intense black color. |
Easier to distinguish between lac + and lac - colonies.
May improve accuracy of automated colony pickers. |
| Complete with LB Agar, S-Gal, IPTG |
No need to make up separate solutions of X-gal and IPTG, saving time and effort. No need to wait for media to cool before adding X-gal and IPTG. No need for toxic solvents, such as DMF. |
| Convenient packaging. Each pouch holds the required amount of S-Gal/LB Agar Blend powder to make up to 500 ml of media. |
Ease of use for the researcher. Just add pre-blended powder to 500 ml of water, autoclave, add antibiotic if desired, and pour plates. Saves time and labor. |
| S-Gal is not heat labile. |
Can be autoclaved or microwaved. Provides flexibility to user. |
| S-Gal is not light sensitive. |
No need to shield product or plates from light. Consistent performance and reliable results. |
Just add water to S-Gal/LB Agar Blend, autoclave and pour your plates. It's that easy!
