Description: The GenElute™ HP Plasmid Miniprep Kits offer purification of high-quality plasmid DNA in less than 30 minutes. These kits provide the convenience of binding columns that can be used with a vacuum or spin purification format. In addition, no alcohol precipitation is required.

Procedure: An overnight recombinant E. coli culture is harvested by centrifugation and subjected to a modified alkaline-SDS lysis procedure followed by adsorption of the plasmid DNA onto silica in the presence of high salts. Contaminants are removed by a vacuum or spin wash step. Finally, the bound plasmid DNA is eluted in water or Tris-EDTA buffer.
Application: High-quality DNA is ready for immediate use for the most demanding applications, including transfection, restriction digestion, ligation, automated sequencing and PCR.

The table on the left depicts the average yield per preparation obtained by the Sigma GenElute HP Plasmid Miniprep as opposed to kits from Supplier Q and Supplier P. The figure on the right depicts the average time per preparation of the GenElute HP Plasmid Miniprep as compared to kits from Supplier Q and Supplier P. All plasmid DNA was isolated following each manufacturers' recommended protocol.

Capillary DNA sequencing of plasmid DNA purified with the GenElute HP Plasmid Miniprep kit. pCMV-SPORT-βgal (7.9 kb) was prepared from 400 µl overnight culture of E. coli host strain DH5α with the GenElute HP Plasmid Miniprep Kit. A 5 µl sample of the preparation was submitted, with adjustment in DNA concentration, to SeqWright DNA Technology Service (Houston, TX) for sequencing witha T7 sequencing primer and ABI BigDye™ terminator v3.1 on an ABI Prism® 3730 DNA Analyzer. The Phred > 20 score was greater than 800 bases.
