Procedure: An overnight recombinant E. coli culture is harvested by centrifugation and subjected to a modified alkaline-SDS lysis procedure followed by adsorption of the plasmid DNA onto silica in the presence of high salts. Contaminants are removed by a vacuum or spin wash step. Finally, the bound plasmid DNA is eluted in water or Tris-EDTA buffer.
Application: High-quality DNA is ready for immediate use for the most demanding applications, including transfection, restriction digestion, ligation, automated sequencing and PCR.
