Description: GenElute™ Plasmid Miniprep Kit offers a simple, rapid, and cost-effective method for isolating up to 20 µg plasmid DNA from E. coli cultures.

Procedure: Bacterial cells are harvested via centrifugation, subjected to a modified alkaline-SDS lysis procedure and the DNA adsorbed onto silica in the presence of high salts. Contaminants are then removed by a simple wash step. Bound DNA is eluted in water or Tris-EDTA buffer.

Application: Plasmid DNA is ready for immediate use in restriction enzyme digestion (Figure 1) cloning, PCR, transcription, conventional and automated sequencing (Figure 2).

Figure 1. Restriction digestions of pUC19 purified from JM109 using the GenElute Plasmid Miniprep Kit. Lane 1 was an undigested plasmid, lanes 2-4 were cut with Bam HI, Eco RI and Pst I respectively. Lane M was 1 kb ladder (11 fragments from 0.5 to 10 kb). Electrophoresis was on a 1% agarose gel.

Cycle Sequencing was performed using 500 ng of template, a T7 sequencing primer, and ABI Big DyeTM terminator chemistry. Sequencing reactions were resolved on an ABI Prism 377 XL instrument with a 48 cm gel cassette containing 4.5% AutoPAGE Plus acrylamide at 2.4 kV, 48°C for 7 hours.
