RNA Purification

RNA extraction and handling are technically more challenging than DNA manipulation due to the presence of a reactive 2’-OH on RNA ribose residues and to the ubiquitous and stable nature of RNases. Successful RNA extraction requires good laboratory practice including special washing and cleaning procedures and RNase-free technique. Products are available for in situ and in vitro procedures and for removing traces of DNA from isolation yields. Sigma’s Spectrum Plant Total RNA kits for isolating and purifying high-quality total RNA needed for applications such as Northern blots and Reverse Transcriptase-Polymerase Chain Reaction (RT-PCR). The kits provide convenient and rapid processing without using hazardous organic solvents, while separating genomic DNA and removing secondary metabolites such as polyphenolic compounds and polysaccharides from a wide variety of plant species and tissues. Yields depend on the tissue type and developmental stage of the sample. The kits can also recover small RNA molecules such as microRNA, siRNA, tRNA, and 5S rRNA.
