Amplification Plot
Log-fold dilutions of human fibroblast genomic DNA (108 to 103 fg) were assayed in triplicate using LuminoCt qPCR ReadyMix and a dual-labeled fluorescent probe and primers targeting the 18S ribosomal RNA gene. Assays were performed on an ABI 7500 Fast qPCR instrument according to the manufacturers recommended protocol.

Standard Curve
The standard curve was generated on an ABI 7500 Fast instrument by plotting the threshold cycle (Ct; see Amplification Plot above) versus the log of the initial quantity of DNA for each reaction.

How to Switch

Switch in 3 simple steps:

  1. Select kit(s) to match desired detection technology
  2. Create rapid protocol(s) for qPCR assays. Sigma recommends*:
    Initial denaturation:95°C for 20 sec
    40 PCR cycles:95°C for 3 sec
    60°C for 15–30 sec
  3. Ensure primers and probes are compatible with rapid assay protocols
     i. Design Primer sets with amplicons of <200 bp; or
    ii. Use Commercially available primer and probe sets (e.g., TaqMan® Assays)

*Reagents are also compatible with the protocols of the following instruments: ABI 7900HT Fast, ABI 7500 Fast, ABI StepOne, Roche LC 480, Bio-Rad CFX96 and CFX 384, and the Eppendorf Mastercycler ep Realplex.
