The Project Situation

A major food testing company was looking to increase its throughput for the analysis of fatty acids. In this analysis, with a traditional method, the analyst spends a lot of time on the first step: the extraction. The aim of the study  is to decrease sample preparation time for the extraction step. By doing so, the analyst will save time for the development of new methods.

MilliporeSigma's Solution

The Folch method has been conventionally used to extract lipids containing fatty acids from biological samples, using chloroform, methanol, and water to separate lipids from aqueous-soluble compounds. In this procedure, lipids are retained in the lower chloroform layer; whereas, aqueous-soluble compounds are retained in the upper methanol-water layer.

The sample is then centrifuged to achieve uniform separation and the bottom chloroform layer is transferred with a pipette to a new test tube.

The Fatty Acid Extraction Kit from MilliporeSigma shortens this extraction process by eliminating the need to prepare solvents and standards, centrifugation, and pipetting. Once the sample is homogenized and dissolved in the Extraction Solvent supplied in our kit, it is inverted twice and poured into the syringe containing a filter, which preferentially elutes the chloroform layer containing total lipids. A portion of the total lipid extract containing fatty acids can then be trans-esterified for GC-FID analysis.

A Solution tested for 27 different matrices

The food testing company saw significant benefits by converting from the traditional method  to the MAK174 kit, including savings of solvents and time.

The customer has tested about 500 extractions total with our kit, approximately 50 per day.

The table below represents the list of matrices tested with the kit, the time saving, and the volume of solvent used per kit.

After extraction and esterification, the customer diluted the sample with Pentane in different volumes depending on the matrice (FINAL DILUTION).

To ensure a sufficient recovery of the final sample, the initial weight of the matrice needed to be adjusted.
