DNAstable® Blood Protocol for preservation of DNA in whole blood at room temperature (96 well plates)

Product Description

DNAstable Blood matrix is a technology for safe, stable, and convenient storage of whole blood and buffy coat at room temperature (15–25°C). This medium is completely dissolvable and ensures total sample recovery.

Storage/Stability

DNAstable blood Tubes & Plates should be stored dry in their original unopened packaging before use.

Procedure

Sample Drying and Storage
Open plate cover from the 96-well plate.

  • Add up to 25 μl of whole blood or buffy coat into each well containing DNAstable Blood matrix.
  • Mix the sample by placing the DNAstable Blood plate (in the carrier plate) on a microtiter plate shaker, and shake for about 15 min at 600-800 rpm.
  • Place AirPore seal across well-plate.
  • Dry the uncovered sample completely using a vacuum evaporator, such as a Speedvac or Vacufuge at room temperature (15–25°C).
  • Store at room temperature (15-25°C), protected from moisture by either:
         1) Storing in a dry storage cabinet or
         2) Heat-seal the moisture barrier bag, including the dried mixture and desiccant packet.

Note: Drying times may vary depending on the humidity level in the laboratory.

Sample Recovery: Just Add Water

  • Add 100 μl of DNAse-free H2O to each well containing stored DNA sample.
  • Place well-plate on a shaker, and incubate at room temperature with continuous shaking at 500 rpm until the blood is rehydrated.
    Note: Rehydration time is dependent on original blood volume and storage time, and may vary from 15-30 minutes. To avoid cross contamination of samples, do not shake at higher speeds.
  • Transfer the rehydrated mixture to a clean microfuge tube and follow the kit protocol for manual or automated purification of DNA from blood or buffy coat.

Note: If using the QIAamp® DNA blood mini kit, repeat the above steps with another 100 μl of DNAse-free H2O.

Purified DNA samples can be used directly in downstream applications such as:

  • PCR
  • qPCR (see handbook for details on dilution factors)
  • Sequencing
  • STR Analysis
  • Whole Genome Amplification
