General description

Glycan analysis has become an increasingly critical aspect of glycomics and proteomics, due to the role of glycoproteins in cell signaling, cell adhesion, immune response, and disease states. Since they tend to have low spectral activity in both UV and visible light, it is often necessary to label glycans in order to enhance detection. GlycoProfile Glycan Clean-Up Cartridges are designed for the post-labeling cleanup of labelled glycans when using the GlycoProfile 2-AA or 2-AB Labeling Kits. Glycans adsorb to the membrane while excess dye and other hydrophobic contaminants pass through. Mono- and disaccharides have a weak affinity for the cartridge membrane and tend to bind poorly. Purified glycans are subsequently desorbed and ready for further enzymatic digestion or analysis by a number of methods, including HPLC.

Application

GlycoProfile Glycan Clean-Up Cartridge is used for clean up of glycan samples after reductive amination labeling or enzymatic digestion. It has also been used to remove excess dye and hydrophobic contaminants from N-linked glycans. The cartridge is equilibrated with acetonitrile, then loaded with a maximum of 20 μg of a glycan sample. The glycans are bound while excess dye and salts are removed by washing with acetonitrile. The glycans are then eluted with water.

Preparation Note

Store the cartridges at room temperature. The cartridges are stable for at least one year at room temperature, as supplied.

Other Notes

Each cartridge is recommended for single use only. The maximum sample size is 10 μL of glycan containing solution and/or 20 μg of glycan.
Recommended for use with the GlycoProfile 2-AA (Cat. No. PP0530) and 2-AB (Cat. No. PP0520) Labeling Kits.
