Application

MDA-MB-231 has been used to study:
  • the changes in the morphological pattern and gene expression of cells when grown in the presence of sera positive for anti-carbonic anhydrase I (CA I) autoantibodies
  • the effects of normothermic conditioned microwave irradiation on cultured cells using an irradiation system
  • the transendothelial migration rates of the cells under different shear stress levels

Preparation Note

L15+ 2mM Glutamine + 15% Foetal Bovine SerumFBS / FCS.
Split sub-confluent cultures (70-80%) 1:3 to 1:6, i.e. seeding at 3-6x10,000 cells/cm2 using 0.05% Trypsin or Trypsin/EDTA; 37°C. CO2 is not required.

    1. Heat inactivated serum is not required for MDA-MB-231 cells. The recommended culture medium is L15 + 2mM Glutamine + 15% Foetal Bovine Serum (FBS).

    1. Product 92020424-1VL is for the cells themselves, MDA-MB-231 Cell Line human. Product 92020424-DNA-5UG is for the corresponding DNA.

  4. Doubts about complete growth media that I have to use with MDA-MB-231. Is it L15 + 2 mM Glutamine+10% FBS or Can I use DMEM, 10% FBS, PenStrep? Protocol that I have to follow after thawing cells (volume, rpm, V to resuspend, FlaskT25 or T75 and ratio)

    1. It is best to adhere to the guidelines recommended by ECACC, at least for bringing cells out of the freeze. Some end users have successfully transitioned cells to other media, but this should always be a gradual transition over several sub-cultures to ensure robust cell growth and morphology.

      The traditionalists advise avoiding using antibiotics, however, if impeccable aseptic technique is a challenge and busy labs and incubators are common, antibiotics are acceptable. Should the added assurance of Pen/Strep be preferred, see product P0781:

      The T25 flasks would be best for starting up cultures. After the culture is established, end users may prefer the T75 flasks, provided there is adequate cell density.

  5. What medium should I use when I thaw the Cell Line cells?

    1. The medium for each cell line is listed on the product page. It is specific to each cell line. If not visible on the Sigma-Aldrich website product page, the HPA website also contains the same information (www.hpacultures.org.uk). The product number for the cell line is the same on either site.

  7. Why is Vapor Phase Liquid Nitrogen preferred for storage of Cell Line cells?

    1. If ampules are immersed into liquid phase of liquid nitrogen, it increases the risk of the liquid seeping into the vial. This could lead to problems of cross-contamination and increased risk of the ampule exploding when thawed.

  10. How do I handle Cell Line frozen cells upon arrival?

    1 answer
    1. Upon receipt, frozen ampules should be transferred directly to vapor phase liquid nitrogen without delay (-135°C) or liquid phase liquid nitrogen if vapor phase is not available. DO NOT use a -80°C freezer as an alternative; this will result in loss of viability.
