General description

4-Methylumbelliferyl phosphate (4-MUP) is a substrate for alkaline phosphatase which forms the soluble fluorescent substance methylumbelliferone. 4-MUP Liquid Substrate System combines 4-MUP and buffer in a ready-to-use reagent.

Application

4-Methylumbelliferyl phosphate (4-MUP) Liquid Substrate System has been used as a fluorescent substrate for alkaline phosphate in enzyme linked immunosorbent assay (ELISA). It has also been used in secreted embryonic alkaline phosphatase (SEAP) activity assay.

Analysis Note

4-MUP fluorescence can be measured with excitation at 360 nm and emission at 440 nm. The fluorescent end product may also be observed using a UV light source.

  1. Which stop solution is the best option for AP-4MUP ELISA? Please advise.

    1. The M3168 product does not require a stop solution. 4MUP is a fluorescent substrate. Adding a stop solution to 4MUP, such as an acidic solution like 2M Sulfuric Acid, would greatly reduce or eliminate the fluorescence. While stop solutions are typically added to substrates like pNPP, TMB, or OPD, they are not required when working with product M3168. The Product Information Sheet for M3168 can be downloaded from the website, where it mentions that the solution is ready to use and nothing else should be added to it.
