3,3′,5,5′-Tetramethylbenzidine has been used as a substrate in competitive enzyme-linked immunosorbent assay (ELISA) and indirect ELISA.
A 3,3′,5,5′-tetrametilbenzidina (TMB) é um substituto não carcinogênico (teste de Ames negativo) para a benzidina adequado como substrato da peroxidase para uso em procedimentos de ELISA. O substrato produz um produto final solúvel de cor azul-clara que pode ser lido espectrofotometricamente em 370 ou 620-650 nm. A reação de TMB pode ser interrompida com H2SO4 2 M (o que resulta em uma cor amarela) e lida em 450 nm. Um reagente sensível e específico para detecção de sangue, teor de hemoglobina, teor de peroxidases.
