Descrição geral

O azul de tripano (TB), um corante azo aniônico hidrofílico, atravessa apenas as membranas celulares das células mortas, colorindo de azul os tecidos/células mortas.[1] É usado em microscopia quantitativa para uma técnica de coloração de exclusão de corante para diferenciar entre células vivas e mortas de mamíferos.[2]
O azul de triptano é um corante usado para distinguir entre células vivas e mortas. É uma mancha vital que não é absorvida por células viáveis saudáveis, mas que cora as células com uma membrana celular danificada.[3] Com isso, apenas células mortas podem ser contadas. O método é às vezes referido como método de exclusão de corantes.

Aplicação

A solução azul de triptano foi usada:
  • no ensaio de viabilidade celular para contar células viáveis e mortas[4]
  • no ensaio de resgate para contar células viáveis no método de contagem de exclusão de[4]
  • azul de triptano para determinar a curva de proliferação[5]
  • como controle salino e na preparação de solução de lipopolissacarídeo (LPS) para confirmar o sucesso da infusão de LPS[6]
  • no teste de exclusão do corante azul de tripano para determinar a viabilidade das células epiteliais gástricas (GEC)[7]
  • para contar as células GEC usando um hemocitômetro[7]

Nota de preparo

Preparado em cloreto de sódio 0,81% e fosfato de potássio dibásico 0,06%.

  4. Why is Trypan blue, Product T8154, used to determine cell viability?

    1. Trypan Blue is a vital stain recommended for use in estimating the proportion of viable cells in a population. The reactivity of this dye is based on the fact that the chromophore is negatively charged and does not react with the cell unless the membrane is damaged. Staining facilitates the visualization of cell morphology. Live (viable) cells do not take up the dye and dead (non-viable) cells do.

  5. Do the cells need to be washed to remove the culture media prior to staining with Product T8154, Trypan Blue solution?

    1. Trypan Blue has a greater affinity for serum proteins than for cellular protein. If the background is too dark, cells should be pelleted and resuspended in protein-free medium or salt solution prior to counting.

  6. Can Trypan blue, Product T8154, be used to discriminate between apoptotic and necrotic cells?

    1. Trypan blue will stain cells that have disrupted membranes.  It cannot differentiate between permeabilized membranes caused by apoptosis or necrosis.

    1. The trypan blue solution is still good to use.  It can be warmed to dissolve the precipitate or it can be filtered to remove the precipitate.

  8. Why are there red fibers in Product T8154, Trypan blue solution?

    1. A report of fibers in Product T8154, Trypan blue solution is sufficiently common almost to be a characteristic of the product. These fibers are a cosmetic fault rather than one of functionality.  The presence of fibers may be more pronounced following cold storage or shipping.  This product should be stored at room temperature. Filtration to remove the fibers will not affect the use of the product.
