General description

BugBuster® Protein Extraction Reagent is formulated for gentle disruption of Escherichia coli cell wall to liberate active proteins. It provides a simple, rapid, low-cost alternative to mechanical methods such as French Press or sonication for releasing expressed target protein in preparation for purification or other applications. The proprietary formulation utilizes a tris-buffer-based mixture of non-ionic and zwitterionic detergents that is capable of cell wall perforation without denaturing the protein. BugBuster is also useful for the preparation of high-purity inclusion bodies for subsequent purification in instances where expressed proteins are insoluble.

Application

BugBuster® Protein Extraction Reagent has been used:
  • to lyse Escherichia coli BLR (DE3) cells for β-glucosidase (BglB) production and purification[1]
  • as a cell lysis buffer to treat the synechocystis PCC6803 transconjugants and measure the yellow fluorescent protein (YFP) fluorescence[2]
  • in lysis buffer for purification of glutathione S-transferase- heat shock protein family A (Hsp70) member 8 (GST- HSPA8)[3]

Questions

1–4 of 4 Questions
  1. Can this buffer be used to extract insoluble proteins for subsequent Ni-NTA purification? Does it contain EDTA?"

    1 answer
    1. This product removes soluble fragments to isolate the insoluble fraction. It does not solubilize insoluble proteins and does not contain EDTA.

  2. Can this be used to extract membrane proteins?

    1 answer
    1. BugBuster Protein Extraction Reagent is a mixture of non-ionic and zwitterionic detergents optimized for the extraction of soluble proteins. It does not solubilize insoluble proteins in inclusion bodies. BugBuster does not lyse cells but creates openings in the membrane to release proteins, similar to most detergent-based extractions. Traditionally, detergents are used to release membrane proteins into the soluble fraction, although not all membrane proteins will be released. Some may remain in the pellet and require denaturing conditions for release. Highly hydrophobic or membrane-associating proteins are likely to be found in the cell debris pellet. The extraction of membrane proteins using BugBuster has not been directly evaluated. Non-ionic detergents can solubilize some membrane proteins, but the extent to which BugBuster achieves this has not been analyzed. It is expected that some, but not all, membrane proteins would be solubilized using BugBuster, and therefore, its use for membrane protein extraction cannot be guaranteed.

  3. can we use this for protein extarction of P aeruginosa bacteria for Carb NP test

    1 answer
    1. BugBuster® Protein Extraction Reagent is formulated for gentle disruption of E. coli cell wall. It is not tested for use with other organisms. Suitability would have to be determined experimentally.
