Can this be used to extract membrane proteins?
1 answer-
BugBuster Protein Extraction Reagent is a mixture of non-ionic and zwitterionic detergents optimized for the extraction of soluble proteins. It does not solubilize insoluble proteins in inclusion bodies. BugBuster does not lyse cells but creates openings in the membrane to release proteins, similar to most detergent-based extractions. Traditionally, detergents are used to release membrane proteins into the soluble fraction, although not all membrane proteins will be released. Some may remain in the pellet and require denaturing conditions for release. Highly hydrophobic or membrane-associating proteins are likely to be found in the cell debris pellet. The extraction of membrane proteins using BugBuster has not been directly evaluated. Non-ionic detergents can solubilize some membrane proteins, but the extent to which BugBuster achieves this has not been analyzed. It is expected that some, but not all, membrane proteins would be solubilized using BugBuster, and therefore, its use for membrane protein extraction cannot be guaranteed.
