General description

Immobilon-P transfer membrane is recommended for most Western blotting applications, especially proteins greater than 20 kDa. Immobilon-P membrane has excellent protein retention, high physical strength, and broad chemical compatibility, making it ideal for a variety of staining applications and reprobing in immunodetection methods.

Antibody reuse for Western blotting is a common practice for many researchers. The recommended blocking agent for PVDF membranes in a Western blot, including when using Casein, as per ther Immobilon®P Transfer Membrane user guide is using a blocking buffer called Block-CH buffer (Cat. No. WBAVDCH01) or a blocking agent such as bovine serum albumin, casein, or nonfat dry milk in wash buffer at a concentration of 0.5-5% (w/v).

      Regarding the suitability of Casein as a blocking agent for PVDF membranes, in-house testing has been conducted with IPVH00010 PVDF membrane, and Casein has been found to be suitable. However, it is important to note that users often choose the blocking agent that works best for their specific needs. Therefore, if one prefers to use Casein as a blocking agent with PVDF membrane (IPVH00010), it can be employed as long as it does not interfere with the samples and antibodies being used.
