General description

Recombinant murine TNF-α is produced in E. coli and purified by standard chromatographic techniques.
Contents:
5 μg/ml solution in PBS and 1 mg/ml BSA, filtered through a 0.2 μm pore size membrane

Specificity

Mouse mTNF-α is effective on mouse, rat, rabbit, and human cells.
Specific activity/EC50: >4 x 108 U/mg, <0.0025 ng/ml (mTNF-a, NIBSC, interim standard, 88/532), at least the same specific activity (EC50) compared to the indicated standard is guaranteed.

Application

TNF-α causes selective necrosis of murine tumors when injected into tumor-bearing mice. In vitro, human TNF-α has direct cytolytic or cytostatic activity on certain transformed cells. In this context, it acts synergistically with interferon-γ. It has also been used for cell stimulation studies.

Quality

Purity: >95% (SDS-PAGE)
Endotoxin level: <0.1 EU/μg (LAL-test), <10 EU/ml (LAL-test)
Note: 1 EU corresponds to 0.1 ng

Specifications

Primary structure: One polypeptide chain (156 amino acids) identical to natural mTNF-a, but not glycosylated. Glycosylation is not essential for biological activity.

Sequence

Chain Length 156 AA

Unit Definition

EC50 definition: The amount of mTNF-α that is required to mediate half-maximal cytotoxicity (MTT cleavage) with WEHI 164 cells in the presence of actinomycin D (1 unit equals ≤0.0025 ng/ml).
