General description

Methyl cellulose is obtained by etherification of cellulose. This commercial cellulose ether exhibits amphiphilic properties and original physicochemical properties. Methyl cellulose is enzyme resistant, non-digestible, non-allergic, and non-toxic.

Application

Methyl cellulose has been used:
  • as a vehicle for metformin drug during oral gavage
  • as a component of mammary epithelial cell growth medium for mammosphere culture
  • as a supplement in spheroid 3D co-cultures for spheroid invasion assays using hanging drop protocol

Methyl cellulose is used to create semi-solid matrices,[1][2] as a cell culture gel, for culture of colony forming cells[3][4] such as CFU-G; CFU-GM; cells and other cells used in toxicity assays. Low viscosity, not for plaquing assays or cloning.

Biochem/physiol Actions

Methyl cellulose finds applications in various industries such as in the food industry it is used as an emulsifier, texturing agent, thickener, and gelling additive. Methyl cellulose is also used in various cosmetics and personal care products such as hair shampoos, hair styling products, liquid soaps, body washes, lotions and creams, and toothpaste to yield their characteristic thick consistency. In addition, it serves as an additive for pharmaceuticals and is used in the production of capsules for nutritional supplements. Methyl cellulose is used as an alternative to glycerol in a cryopreservation medium. It is used along with culture supplements to enhance proliferation and differentiation in multiple or single lineage-committed hematopoietic progenitor cells. This cellulose derivative also has the potential to prevent the loss of adhesive properties and cell functions of various cell types including human skin fibroblasts, human melanoma cells, and mouse lung fibroblasts cultured in suspension.

  1. Which product is suitable for spheroid culture and How can I prepare DMEM media with 0,24% (w/v) for spheroid culture?

    1. The methylcellulose products available are not currently tested for use in spheroid culture. However, a literature search found that item M0512 has been used for culturing spheroids. Here are references for the preparation of methylcellulose in media:

  2. How to practically reach a 1% solution of Methylcellulose in cell culture media that can't be heated excessively?

    1. This product is supplied in powder form. The solubility in water is 50 mg/mL, resulting in a hazy solution. To prepare a 1% Methylcellulose solution in cell culture media from this stock solution, follow these steps:
      A 1% solution requires 1 g of solute per 100 mL of solution, equivalent to 10 mg/mL. The stock solution is 50 mg/mL. To achieve this, take 20 mL of the 50 mg/mL stock solution and dilute it to a total volume of 100 mL.

  3. I would like to use methylcellulose in my cell culture media to optimize organoids in suspension. What methylcellulose product would be the most appropriate? And can I dissolve it directly into the cold media?

    1. The methylcellulose products available are not currently tested for use in organoid suspension. However, a previously available product, S3812, was an IMDM-based stem cell culture medium that included a methylcellulose component with a 4000 cps viscosity, at a concentration of 1%. This would correspond to product M0512. The solubility in cold medium has not been determined.   4. Can the M7027 pass through a 0.2um filter?

    1. Methyl cellulose has a high molecular weight, and the viscosity of methyl cellulose solutions can also impede its ability to pass through such fine filters, as it can clog the filter pores. For sterilization, a 0.5-1% solution of methylcellulose can be autoclaved using standard conditions (121 °C, 15 psi, 15-20 minutes). Higher concentrations may lead to a portion of the methylcellulose falling out of solution.

  5. Does this product need to be sterilized after dissolving for use in cell culture?

    1. Solutions of this material require sterilization for cell culture applications. A 0.5-1% solution of methylcellulose can be autoclaved using standard conditions (121 °C, 15 psi, 15-20 minutes). Higher concentrations may lead to a portion of the methylcellulose falling out of the solution.

    1. The molecular weight is not analyzed on a lot to lot basis. The approximate molecular weight is 88,000 Da with methoxy substitution between 27.5 - 31.5% (weight).

  7. Can I use this to make 0.5% solution in water which will be used as the solvent to dissolve neratinib and administrate to mice through oral gavage?

    1. This product can be solubilized in water at 50mg/ml which is a 0.5% solution. This product is tested for use in cell cultue, however it has not been tested for in vivo applications. Suitability for in vivo use will need to be validated by the end-user. See the link below for a general product information sheet for Methyl Cellulose. Note product M7140, which is the general research grade of this product. All details for M7140 will apply to M7027.
