General description

IL15 (interleukin 15) gene is localized to human chromosome 4q31, composed of eight exons, and the encoded protein is a short-chain α-helical bundle type 1 cytokine. IL15, along with its receptor IL15Rα, are co-expressed majorly by activated monocytes and DCs (dendritic cells).

Biochem/physiol Actions

IL15 (interleukin 15) and IL2 are the only two known cytokines till date that bind to T-cells through IL-2Rβ (IL2 receptor β). It interacts with lymphocytes via the heterotrimeric IL-15 receptor (IL-15R) composed of IL-15Rα, IL-2 receptor β (IL-2Rβ), and IL-2 receptor γ (IL-2Rγ). Since, IL-2R and IL-15R have common subunits, it is supposed that IL2 and IL15 perform similar functions, such as induction of lymphocyte activation and proliferation in vitro, and apoptosis of activated lymphocytes (AICD). However, unlike IL2, IL15 mimics the function of IFN I (interferon) by potently and selectively activating memory-phenotype CD44hi CD8+ cells.

Physical form

Lyophilized from 0.22 μm filtered solution in 20 mM Tris, 100 mM NaCl, pH 8.0. Generally 5-8% Mannitol or trehalose is added as a protectant before lyophilization.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in sterile deionized water to a concentration of 100 μg/mL. Do not vortex. This solution can be stored at 2-8°C for up to 1 month. For extended storage, it is recommended to store at -20°C.
