Specificity

Clone 1K9 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects CCR6/CD196. It targets an epitope within 16 amino acids from the third extracellular domain.

Immunogen

KLH-conjugated linear peptide corresponding to 16 amino acids from an extracellular domain of human CCR6/CD196.

Application

Quality Control Testing

Evaluated by Western Blotting in HuH-7 cell lysate.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected CCR6/CD196 in HuH-7 cell lysate.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected CCR6/CD196 in human spleen tissue sections.

Flow Cytometry Analysis: 1 µg from a representative lot detected CCR6/CD196 in human peripheral blood mononuclear cells (PBMC).

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected CCR6/CD196 in HL-60 cells.

Affinity Binding Assay: A representative lot of this antibody bound CCR6/CD196 peptide with at least one hundred thousand-fold (100,000X) higher than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

C-C chemokine receptor type 6 (UniProt: P51684; also known as C-C CKR-6, CC-CKR-6, CCR-6, Chemokine receptor-like 3, CKR-L3, DRY6, G-protein coupled receptor 29, GPR-CY4, GPRCY4, LARC receptor) is encoded by the CCR6 (also known as CKRL3, CMKBR6, GPR29, STRL22) gene (Gene ID: 1235) in human. The gene is mapped to human chromosome 6q27. It shares sequence similarity with G protein-coupled chemokine receptor. CCR6 plays a crucial role in the immune system by mediating the migration and positioning of various immune cells. CCR6 encodes a chemokine receptor that is mainly expressed in B cells and memory T cells. It is also found to be expressed in dendritic cells, spleen, thymus, small intestine, and peripheral blood leukocytes. It is primarily localized on the plasma membrane of cells, where it interacts with its ligand CCL20. CCL20 is the only known high-affinity ligand that binds to CCR6. Upon ligand binding, CCR6 undergoes conformational changes that activate intracellular signaling pathways, leading to cellular responses such as chemotaxis. CCR6 has been implicated in various diseases, particularly those involving inflammatory and autoimmune responses. Overexpression of CCR6 has been observed in certain cancers, such as colorectal cancer and hepatocellular carcinoma. Studies show a very strong correlation between the expression of CCR6 and disease severity in patients with inflammatory bowel disease (IBD), psoriasis (PS), rheumatoid arthritis (RA), and multiple sclerosis (MS). CCR6 could be a potential therapeutic target in these diseases and, CCR6 antibodies could be a promising alternative to small molecules in the treatment of CCR6/CCL20 axis-related pathologies. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
