Description

Anwendung

Das Entfernen von Proteinen aus Nukleinsäuren kann durch Extraktion mit Phenol/Chloroform/Isoamylalkohollösungen erzielt werden. Für Rohgemische aus Nukleinsäuren kann es nützlich sein, vor der Phenol/Chloroform-Extraktion einen Verdau mit einem vielseitig anwendbaren proteolytischen Enzym vorzunehmen. Obwohl Proteine effizient durch Phenol denaturiert werden, wird die RNase-Aktivität nicht völlig gehemmt. Daher wird eine kleine Menge Isoamyl-Alkohol hinzugefügt, um die Deaktivierung der RNase-Aktivität zusätzlich sicherzustellen. Warnhinweis

Kommen Phenollösungen mit der Haut in Kontakt, kommt es schnell zu Verbrennungen. Der Anwender muss daher angemessene Laborkleidung einschließlich Handschuhe und Schutzbrille tragen. Bei direktem Hautkontakt mit viel Wasser spülen und ggf. einen Arzt hinzuziehen.

Für die DNA-Extraktion kann der pH-Wert der Phenolphase durch Equilibrierung mit Tris-Puffer auf 8,0 angepasst werden. Sigma bietet Produkt P2069 speziell für DNA-Extraktionen an. Dieses Produkt enthält Equilibrierungspuffer, um den pH-Wert anpassen zu können.
