General description

Specificity

Clone 1H10 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects PIEZO2. It targets an epitope within 20 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the C-terminal half of human PIEZO2.

Application

Anti-PIEZO2, clone 1H10 ZooMAb®, Cat. No. ZRB1468, is a recombinant Rabbit monoclonal antibody that detects PIEZO2 and is tested for use in Affinity Binding Assay, Immunocytochemistry, and Western Blotting.
Quality Control Testing

Evaluated by Western Blotting in HepG2 cell lysate.

Western Blotting Analysis: 1:10,000 dilution of this antibody detected PIEZO2 in HepG2 cell lysate.

Tested applications

Western Blotting Analysis: 1:10,000 dilution from a representative lot detected PIEZO2 in HeLa, NIH3T3, and C6 cell lysates.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PIEZO2 in HeLa, A431, and NIH3T3 cells.

Affinity Binding Assay: A representative lot of this antibody bound PIEZO2 with a KD of 5.1 x 10-7 in an affinity binding assay.

Target description

Piezo-type mechanosensitive ion channel component 2 (UniProt: Q9H5I5; also known as Protein FAM38B) is encoded by the PIEZO2 (also known as C18orf30, C18orf58, FAM38B) gene (Gene ID: 63895) in human. PIEZO1 and PIEZO2 are mechanically activated ion channels that confer mechanosensitivity to a wide variety of cell types. They differ in their physiological roles and biophysical properties. PIEZO1 is reported to mediate shear stress and stretch-induced transmembrane currents in nonneuronal cells and PIEZO2 is predominantly expressed in sensory neurons and is required for proprioception , the detection of light touch, and the detection of painful mechanical stimuli. PIEZO2 is a multi-pass membrane glycoprotein with 37 transmembrane domains. Mutations in PIEZO2 gene are known to cause distal Arthogryposis that is characterized by congenital joint contractures that mainly involve two or more distal parts of the limbs, in the absence of a primary neurological or muscle disease. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
