General description

Phenol:Chloroform:Iso-amyl alcohol (125:24:1) is used for the purification of total RNA from mixtures of crude cell extracts and can eliminate the need for a time consuming ultracentrifugation step. The addition of phenol, chloroform, and isoamyl can be employed to aid in the separation of lipids and cellular debris, leading to the transfer of these components to the organic phase. Meanwhile, the genomic DNA (gDNA) remains in the aqueous phase, effectively isolating it from the other substances.[1]

Application

Phenol:Chloroform:Iso-amyl alcohol (125:24:1) has been used:
  • for the extraction and purification of RNA.[2][3][4]
  • to isolate RNA from Arabidopsis siliques.[5]
  • to extract proteins and DNA from polysomal and non-polysomal fractions.[6]
  • in TRIzol methods to prepare and extract miRNAs from cells, tissue, and whole flies.[7]
