General description

Alzheimer′s disease (AD) is a progressive neurodegenerative disorder and the leading cause of dementia, affecting approximately 36 million people worldwide. The disease is characterized by two key features: extracellular amyloid β (Aβ) plaques formed from peptides like Aβ1-40 and Aβ1-42, and intracellular neurofibrillary tangles composed of hyperphosphorylated Tau protein. These biomarkers—reduced soluble Aβ1-42, increased total Tau, and phosphorylated Tau in cerebrospinal fluid—serve as critical indicators of the pathological processes underlying neurodegenerative conditions.

  1. Can you provide information on the specificity or cross reactivity of Total Tau in the human HNABTMAG-68K?

    1. The human panel HNABTMAG-68k follows the Tau standard with isoform Tau441. Both the capture and detection immunogens used are also Tau441. Cross reactivity for other isoforms has not been tested.

  2. Can the Human Amyoid Beta / Tau (HNABTMAG-68K) Milliplex kit, which currently only lists CSF as a sample type, be used with serum/plasma samples?

    1. The HNABTMAG-68K Milliplex kit currently only lists cerebrospinal fluid (CSF) as a sample type. When serum/plasma samples were tested with this kit, a strong matrix effect was observed. As the kit was not validated for serum/plasma, the appropriate Serum Matrix for use could not be ascertained. Furthermore, the expected trend for Amyloid β40 and Amyloid β42 in serum/plasma was not observed.

      For serum/plasma samples, we recommend using the validated ELISA kits for Amyloid β40 and 42 (EZHS and EZH42) as they are the preferred assays. Additionally, we offer a Total Tau ELISA (RAB1085).

  3. Are the antibodies used in the Human Amyloid Beta/Tau (HNABTMAG-68K) kit also able to recognize rat samples?

    1. The AB40 and AB42 antibodies have not been specifically tested for reactivity to rat. However, in a test involving 4 rat serum samples, AB40 was detected in all of the samples, while AB42 was not detectable. Additionally, the capture antibodies for pTau and Total Tau have been found to be reactive to both human and rat, as pTau and Total Tau were detected in 2 out of the 4 rat serum samples tested. It's important to note that the panel has not been tested with rat standards to determine the extent of cross-reactivity.

  4. What is the recommended concentration of guanidine for Amyloid Beta Immunoassays?

    1. The recommended guanidine concentration for Amyloid Beta Immunoassays should be less than 0.5M in the final sample concentration.

  5. Could you provide the plate capacity for an LX200 or FM3D instrument when using one box of Sheath Fluid?

    1 answer
    1. The FLEXMAP 3D and LX200 instruments use approximately 10 mL of sheath per sample run when running a 96-well plate and the xPONENT protocol is set to aspirate 50 µL per sample. Therefore, to acquire one 96-well plate, the instruments will use approximately 1L of sheath fluid.

  6. Is it possible to add other analytes to the Human Amyloid Beta / Tau Panel (HNABTMAG-68K) as SPR kits?

    1. Additional analytes cannot be added to the AB/Tau panel due to differences in the assay procedure. The detection antibody is added at the primary incubation, whereas the other panels have the detection added after the primary incubation.

  7. How should the plate for the complete Milliplex assay in Sheath Fluid be stored if it can't be read for a couple of hours?

    1. The plate can be stored at 4°C without shaking for up to 24 hours. Before reading, the plate should be shaken at room temperature for at least 20 minutes to bring it back to ambient temperature.
