General description

Die Imprint™ methylierte DNA-Quantifizierungstechnologie bietet eine schnelle und verlässliche Methode zum Messen von globalen DNA-Methylierungsverschiebungen. Das Imprint methylierte DNA-Quantifizierungskit enthält alle Reagenzien, die zum Nachweis von relativen Mengen methylierter DNA erforderlich sind. Bis zu 200 ng aufgereinigte DNA werden an die Wells des Assay-Streifens gebunden. Die methylierte DNA wird durch die Einfang- und Nachweis-Antikörper nachgewiesen und anschließend kolorimetrisch quantifiziert. Die in der Probe vorhandene Menge von methylierter DNA ist proportional zu der gemessenen Absorption.

  1. Could you please provide the sensitivity of the kit and explain what differences in methylation status it can detect?

    1. Based on previous results, a 20% difference in methylation is typically observable. It is recommended to conduct replicate experiments, assess the margin of error (technical and/or biological), and utilize a p-test or other statistical approach to evaluate its applicability in your specific case.

    1. The antibody works on 5-me-C, so any species with DNA that is methylated at the 5 position of cytidine residues will work.

    1. No, you can use as many wells as needed.  You will want to ensure that all the wells that are not being used are covered well with Parafilm, Saran Wrap or some other plate sealer.

    1. No. A fresh dilution of the antibody should be prepared each time the kit is used. Diluting the antibody all at once will decrease its stability and it's effectiveness in the kit.

  5. How does the storage temperature relate to shipping conditions?

    1. The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.
