General description

Amicon® Ultra-0.5 centrifugal filter devices provide fast ultrafiltration, with the capability for high concentration factors and easy concentrate recovery from dilute and complex sample matrices. For example, Ovalbumin (45 kDa) at 1 mg/ml initial concentration is typically retained at >95% .

The vertical design and available membrane surface area provide fast sample processing, high sample recovery (typically greater than 90% of dilute starting solution), and the capability for 30-fold concentration. Solute polarization and subsequent fouling of the membrane are minimized by the vertical design, and a physical deadstop in the filter device prevents spinning to dryness and potential sample loss. Efficient recovery of the concentrated sample (retained species) is achieved by a convenient reverse spin step after collecting the filtrate.

Amicon® Ultra-0.5 devices are supplied non-sterile and are for single use only. With their vertical membrane design and reverse spin, Amicon® Ultra 0.5 mL filters deliver great performance and lower spin times.
We are committed to bringing you Greener Alternative Products, which have been re-engineered to enable more sustainable research solutions. The inner filtration device is made from bio-circular material allocated using the ISCC mass balance approach. Click here for more information.

Protocols

Detergents are frequently used to solubilize proteins and nucleic acids during purification, but the presence of detergents may interfere with downstream analyses.

If an antibody must be purified from a small sample volume (0.5-10 mL), then the Amicon® Pro Purification System is recommended.

Articles

PNGase Fast denaturing buffer and enzyme provide results similar to a conventional 20-hour protocol, reducing workflow time to about 1 hour.

Commercially available protein labeling kits provide researchers with the flexibility to customize their own immunoassay detection panels.

Protein purification is simpler with Amicon Pro. The device delivers consistent sample preparation for more reliable recovery, uncompromised purity and easier data generation.

Questions

1–4 of 4 Questions
  1. What is the maximum Urea´s concentration that can resisten this Amicon Ultra 30 KDa?

    1 answer
    1. From the compatibility chart in the user guide, the devices can tolerate less than or equal to 8M urea.

      Helpful?

  2. What is the maximum speed of an Amicon Ultra 2 device?

    1 answer
    1. The maximum speeds for concentration spins as per the User Guide are as follows:
      • Swinging bucket rotor: 4,000 × g
      • Fixed angle rotor: 7,500 × g
It is important to note that when spinning viscous solutions like undiluted serum or plasma, the maximum speed should not exceed 5,400 x g.

      Helpful?

  3. what is the pore sizes of 30kDa, 50kDA, and 100 kDa filters? And their corresponding maximum and minimum volume which can be used?

    1 answer
    1. These ultrafiltration do not have defined pore sizes. They are characterized by retention of globular proteins, and assigned nominal pore ratings. These MWCO membrane types are found in centrifugal devices with 70, 15, 4, 2, and 0.5 ml volume capacities.

      Helpful?

  4. Can I use Amicon® Ultra Centrifugal Filter, 30 kDa MWCO to measure human serum protein binding for a small moelcule?

    1 answer
    1. The Centrifree® Ultrafiltration Centrifugal Filters are recommended for this application over the Amicon® Ultra Centrifugal Filters. Please see the link below:
