Cell-permeable phosphatidylinositol 3-kinase (PI 3-kinase, PI3K) activator.
740 Y-P is a cell-permeable phosphatidylinositol 3-kinase (PI 3-kinase, PI3K) activator composed of the Drosophila antennapedia-derived membrane-permeant 16-mer sequence RQIKIWFQNRRMKWKK followed by a phosphotyrosine-containing, PDGFR-derived sequence SDGGY(P)MDMS that targets SH2 domains of PI3K p85 regulatory subunit. When applied to serum-starved C2 cells at 50 µg/mL, 740 Y-P is more effective than 10% FCS, 5 ng/mL bFGF or 10 ng/mL EGF in stimulating cell proliferation (% of cells in S-phase post 48h stimulation = 48.3, 42.5, 35.7 and 32.8, respectively). 740 Y-P-induced mitogenic response is blocked by wortmannin or rapamycin, but not MEK inhibitor PD98058.
