Application

Urease from Canavalia ensiformis (Jack bean) has been used: to spike urine samples to determine the hydrolysis rate of urea in pure urine[1] It has also been used to study the inhibitory action of polyhalogenated benzo- and naphthoquinones on Bacillus pasteurii and Canavalia ensiformis.[2]

Biochem/physiol Actions

In Canavalia ensiformis (Jack Bean), urease facilitates the conversion of L-canavanine to canaline. [3]
Urease is involved in purine metabolism and the urea cycle. It hydrolyze urea to produce ammonia and carbon dioxide.[4]

General description

Subunit molecular weight: ~90,770
Composed of six subunits with total molecular weight: ~544,620
Urease (Ure) enzyme is present in the cytosol. It is active at acidic pH and is expressed at 28°C.[5] Urease binds to nickel and is present in archaea, bacteria, unicellular eukaryotes and plants.[6] Jack bean urease was the first enzyme to be crystallized and the first enzyme found to contain nickel.[7] It is a multi-subunit enzyme, consisting of 91 kDa subunits in three protein forms.

Other Notes

One micromolar unit will liberate 1.0 μmole of NH3 from urea per min at pH 7.0 at 25 °C. It is equivalent to 1.0 I.U. or 0.054 Sumner unit (1.0 mg ammonia nitrogen in 5 minutes at pH 7.0 at 20 °C)

  1. Hi there I was wondering what Type IX means and how many grams are there in the 20 kU one?

    1. The 'type' is a historical designation used to differentiate the various Urease products and generally indicates a specific method of isolation and/or purification.  The Type IX undergoes dialysis against deionized water prior to lyophilization.  The process is EDTA-free, but may leave traces of potassium phosphate buffer salts.  This item has an activity range of 50000 - 100000 units per gram solid. The mass of the 20KU option may vary from 200 mg – 400 mg per vial.  The lot specific activity is published in the Certificate of Analysis.

    1. The type designations for urease products are not indications of particular isoforms. They are internal codes that refer to the chronological order of the products' market entry and manufacturing processes. The manufacturing processes are proprietary. However, the major differences between the urease products are their specifications for specific activity. Please access the 'Specification Sheet' and 'Sample Certificate of Analysis' for each individual product via its corresponding website.

    1. The 'type' is a historical designation used to differentiate various Urease products. Each lot has a specific activity of 50,000 - 100,000 units per gram solid. Therefore, the mass of the 20 kU option can vary between 0.2 g and 0.4 g. The lot-specific certificate of analysis will provide the specific activity.
