应用

分散酶® 用于制备来自多种不同组织和器官的细胞。 该酶已被证明是一种快速、有效但温和的试剂,用于破坏组织的细胞外基质,以释放单细胞用于细胞培养(原代细胞培养)或收获已在培养物中的细胞以转移至新基质(二次培养)。此外,其还可用于防止悬浮培养细胞的不必要聚集。

1 个单位的罗氏应用分散酶®等于 181 个蛋白酶单位 (PU),以 pH 7.5 和 + 37°C 下相当于每分钟每毫升 1 μg 酪氨酸的氨基酸释放量测量。

制备说明

激活剂:最适 Ca2+ 浓度为 2 mm,酶制剂中含有足够的 Ca2+,以达到最佳活性。
工作浓度: 0.6~2.4 U/mL
工作溶液:贮备液和工作溶液的制备:
为制备 10 mg/mL 贮备液,将冻干分散酶® I 酶溶于 HEPES 缓冲盐水 (50 mM HEPES/KOH pH 7.4,150 mM NaCl) 中。为制备工作液,用分离细胞的培养基稀释上述贮备液,最终浓度为 0.6-2.4 U/mL。请注意,不建议浓度高于 2.4 U/mL。为获得最佳结果,使用 0.22μm 过滤器膜。
储存条件(工作溶液):-15°C 至-25°C
复溶贮备液在 2 ~ 8°C 下可稳定保存 2 周。储存长达 2 个月时,贮备液应等分冷冻。避免反复冻融。
用 PBS 稀释的工作液在 2~8°C 可稳定保存 3 天。
