一般描述

牛胰腺脱氧核糖核酸酶I(DNase I)是DNA小沟相互作用的核酸酶,特异性相对较低。该蛋白质由两个中心β 折叠构成,而每个折叠由六个β-链所组成。该结构被大量的环以及α-螺旋区域包围。该酶与核酸外切酶III具有结构相似性。DNase I是研究得最透彻的哺乳动物来源的内切酶之一。它是一种双链特异性内切核酸酶,需要二价阳离子激发最大活性。

应用

蛋白质(膜蛋白)的分离操作。
牛胰腺脱氧核糖核酸酶I(DNase I)已被用于:
  • 分离肺、皮肤和肿瘤样品中的细胞
  • 从抗DNase I的活细菌细胞中提取细菌DNA †

制备说明

激活剂:Ca2+ (0.12 mM)与Mg2+结合在一起可增强活性 [Laskowski (1971); Melgar及Goldthwait (1968)].
工作浓度:1 mg/ml
在溶解过程中请勿涡旋!
储备条件 (工作溶液):当在水中重悬时,溶液可在2至8 °C下保存2至3天。在溶解过程中请勿涡旋。注:至少以1 mg/ml溶解。
