一般描述

胶原酶是一种蛋白酶,可切割三螺旋结构的蛋白,即胶原蛋白。溶组织梭菌 产生六种类型的胶原酶,即α、β、γ、δ、ε和λ。这些胶原酶对结缔组织具有活性,并且对明胶的反应性高于胶原。来自溶组织梭菌( Clostridium histolyticum )的胶原酶具有非常强的活性,它在低至4-10°C的温度下可从两端消化胶原蛋白。

特异性

胶原酶可降解天然胶原蛋白。梭菌蛋白酶,一种胰蛋白酶样酶,以及中性蛋白酶也会降解其他蛋白质。

应用

来自溶组织梭菌( Clostridium histolyticum )的胶原酶被广泛用于许多组织类型(例如,肺、心脏、肌肉、骨骼、脂肪组织、肝脏、肾脏、软骨、乳腺、胎盘、血管、脑、肿瘤和脐带等)的解离,并用于制备单细胞悬浮液以便建立原代细胞培养系统。当产量、活力和功能性很重要时,推荐使用胶原酶A。
Roche公司的梭菌 原酶已被用于制备来自多种组织的细胞,如肝细胞、脂肪细胞、胰岛细胞、上皮细胞、肌细胞、内皮细胞 。但是,应根据经验确定各批次的酶是否适用于特定组织的解离。

单位定义

单位定义:来自Roche的胶原酶以Wünsch单位表示(在+25°C 下,每分钟使用Wünsch底物生成1 μmol产物)。
通常,胶原酶活性以Mandl单位表示(在+37°C下,在5小时内从胶原中释放1 μmol亮氨酸)。
遗憾的是,两种活性单位之间没有一致的换算因数,因为Mandl单位部分取决于胶原酶制剂中污染蛋白酶的浓度,而这是一个不确定变量。实际上,高纯度胶原酶制剂的Mandl单位比酶活要低于粗制胶原酶制剂。梭菌 制剂的换算因数通常约为1:1800(例如 ,指定批次的 梭菌 胶原酶含有约0.15 Wünsch U/mg和250 Mandl U/mg)。

罗氏建议仅重新配立即使用所需的冻干粉的量。重新配制的溶液可以在-15至-25 °C下储存长达一周。因为重新配制后活性降低了,所以要避免反复冻融。
