一般描述

分散酶是一种中性、快速反应、温和的蛋白酶,能将表皮从真皮中完整分离。它还能将培养的上皮细胞从基质中完整分离出来。其可分裂基底膜区,使上皮细胞保持完整。胶原酶是一种分解三螺旋蛋白胶原蛋白的蛋白酶。组织胶原酶分三种类型,它们属于基质金属蛋白酶(MMP)家族。从溶组织性梭状芽孢杆菌中提取的胶原酶具有很强的活性,因为它可在低至4-10℃的温度下,从两端消化胶原蛋白。

特异性

胶原酶降解天然胶原蛋白。中性蛋白酶® 是一种非特异性的金属蛋白酶。

应用

胶原酶/中性蛋白酶® 用于从多种不同的组织和器官中制备细胞。

组分

EC 3.4.24.4 & 3.4.24.3

单位定义

一个单位是指,在1分钟内在反应条件下释放出1 μmol酪氨酸对应的Folin-阳性氨基酸和多肽所需的酶活性。

制备说明

激活剂:Ca2+

工作浓度:约为1mg/mL(胶原酶:0.1U/mL,中性蛋白酶:0.8U/mL

酶活性:
胶原酶:0.1 Wünsch U/mg冻干粉(+ 25°C,4-苯基-偶氮苄基-氧基羰基-Pro-Leu-Gly-Pro-D-Arg作为底物)
中性蛋白酶®:0.8 Wünsch U/mg冻干粉(+ 37°C,酪蛋白为底物,pH 7.5)
污染蛋白水解活性:<0.1U/mg 冻干粉 (血红蛋白作为底物)

工作溶液:工作溶液制备 将酶溶于水中,使其终浓度为100mg/mL。用PBS(不含Ca2+/Mg2+)稀释至终浓度为1mg/mL,并采用无菌0.2μm 过滤膜过滤。
吴军溶液的酶浓度:胶原酶,0.1U/mL PBS; 中性蛋白酶®, 0.8U/mL PBS。

抑制剂:
胶原酶抑制剂:EDTA, EGTA
血清不能抑制胶原酶/中性蛋白酶。

保存条件(工作溶液):-15 至 -25°C
复溶的储备溶液在-15至-25°C下稳定。
注:适当分装保存并避免反复冻融!

重悬

复溶于水中(100mg/mL)后续稀释采用PBS(磷酸缓冲盐)(不含Ca2+/Mg2+)。
