牛胰腺核糖核酸酶A是核糖核酸酶A蛋白质超家族的成员之一。它是最具有特征的蛋白质,是肾脏形状的基本蛋白质。该蛋白质由124个氨基酸组成。核糖核酸酶A的天然形式是同二聚体。

生化/生理作用

核糖核酸酶A催化RNA转磷酸化和降解。它是第一种被确认具有使DNA变性功能的蛋白质。该酶能够和单链(ss)RNA聚核苷酸链上多个结合位点结合,然后降解它。它和身上DNA以相似的方式发生作用,导致DNA解旋。通过串联获得的核糖核酸酶A二聚体对癌症细胞有毒性。

单位定义

一单位酶活性导致A0比A1吸收率在实验条件下降低。A0 比A1对应总转化,A1是终吸收率。

重悬

核糖核酸酶A在10mM Tris-HCl,pH7.5,15mM NaCl中溶解,浓度为1到10mg/ml。加热到100℃,15分钟使得里面的脱氧核糖核酸酶失活,然后逐渐降温到室温,分装。Roche建议随后储存在-15到-25℃。
