一般描述

SIRT1 检测试剂盒提供测量纯化 SIRT1 活性和筛选抑制剂/活化剂所需的所有试剂。
测定程序基于两步酶促反应。第一步是通过 SIRT1 对含有乙酰化赖氨酸侧链的底物进行脱乙酰化。第二步是通过显影溶液裂解脱乙酰底物，并释放高度荧光基团。测量的荧光与样品中酶的脱乙酰活性成正比。

应用

SIRT1检测试剂盒(MSCs)用于检测间充质干细胞(MSC)、人视网膜内皮细胞(HREC)和人神经母细胞瘤SH-SY5Y细胞中的Sirt1去乙酰化酶活性。

生化/生理作用

去乙酰化酶（Sir2）是 NAD+ 依赖性组蛋白/蛋白质脱乙酰酶的进化保守家族，其紧密结合 NAD+ 的裂解和蛋白质底物的脱乙酰化。反应产物是烟酰胺、脱乙酰产物和新的代谢物，2′-O-乙酰基-ADP-核糖。该家族中的蛋白质以从该家族发现的第一种蛋白质 Sir2（Silent Information Regulator 2，沉默信息调节因子 2）命名。除了基因沉默外，去乙酰化酶蛋白在其他过程中也很重要，如细胞周期调节和脂肪酸代谢。SIRT1 是 Sir2 的人类同源蛋白，也是迄今为止研究最多的一种。SIRT1 介导 p53 依赖性过程，转录调节、肌肉分化、脂肪形成和轴突变性保护。SIRT1 还参与早期胚胎发生、神经发生和心脏发生。
