牛脱氧核糖核酸酶I已用于一项比较脾脏脱氧核糖核酸酶和胰脱氧核糖核酸酶引发作用的研究中。牛脱氧核糖核酸酶I也已用于研究脱氧胸苷3′、5′-对-对硝基苯基磷酸盐作为牛胰脱氧核糖核酸酶的合成底物。

生化/生理作用

DNase I是一种内切核酸酶,可作用于磷酸二酯键(与嘧啶相邻)以产生具有末端5′-磷酸的多核苷酸。其最适pH在7和8之间。二价阳离子如Mn2+、Ca2+、Co2+和Zn2+都是酶的活化剂。5 mM浓度的Ca2+可稳定酶免收蛋白水解酶的降解。已知2-巯基乙醇、螯合剂、十二烷基硫酸钠(SDS)和肌动蛋白都可抑制酶的活性。
将单链和双链 DNA 消化至携带 5' 的单链和寡核苷酸混合物中磷酸盐和 3′ OH终点。这种催化活性是二价离子依赖性的。在 M 2+ 存在下,DNaseⅠ 随机独立地水解双链 DNA 各链。在 Mn2+ 存在下,两条链均可裂解。
