应用

用适当的过氧化物酶底物检测含有β(1→4)-N-乙酰基-D-葡糖胺的糖蛋白
通用Western Blot实验方法:
  • 糖蛋白样品规格:500ng
  • 凝集素浓度:0.1ug/ml

  1. 每条泳道上样500 ng糖蛋白样品
  2. 进行4-20% Bis-Tris SDS page凝胶
  3. 转膜至PVDF膜上
  4. 用于RIPA缓冲液(R0278 Sigma)在室温对膜进行封闭1小时
  5. 在室温用RIPA缓冲液以0.1 ug/ml孵育HRP凝集素2小时
  6. 用25ml RIPA缓冲液洗膜 5 x 5分钟
  7. 用化学发光底物(CPS1-120)进行检测

生化/生理作用

WGA 不具有血型特异性,但具有与 N-乙酰基-β-D-葡糖胺残基和 N-乙酰基-β-D-葡糖胺低聚物的亲和性。WGA 不含蛋白结合糖。

制备说明

由亲和纯化凝集素制备偶联物。
通过改良的O′Sullivan方法,使用VI型过氧化物酶(P8375)而制备,对低分子量耦合物具有倾向性。在耦合后通过亲和层析进行再纯化。
