该产品是来自大肠杆菌血清型 O26:B6的酚提取物。源菌株是ATCC 12795。来自血清型 026的LPS:B6是来自大肠杆菌的唯一 LPS,其在SDS-PAGE 上表现出短链长行为。该 LPS的短链长度接近突变粗糙株 LPS的短链长。该 LPS 血清型(1ug/ml),用于刺激中性粒细胞。

应用

脂多糖(LPS)是革兰氏阴性细菌细胞壁的特征成分。LPS及其脂质A部分通过Toll样受体4(TLR4)刺激先天免疫系统的细胞,TLR4是Toll样受体蛋白家族的成员,该家族可识别常见的病原体相关分子模式(PAMP)。

生化/生理作用

脂多糖(LPS)位于膜的外层,在非包封的菌株中,暴露在细胞表面上。它们有助于外膜的完整性,并保护细胞免受胆汁盐和亲脂性抗生素的影响。

制备说明

产物可溶于水(5mg/ml)或细胞培养基(1mg/ml),产生浑浊的、淡黄色溶液。在经过涡旋和加温至 70-80oC 后,在水溶的盐水中得到更浓缩、但仍然浑浊的溶液(20mg/ml)。脂多糖是在各种溶剂中形成胶束的分子。在水和磷酸盐缓冲盐水中观察到浑浊的溶液。有机溶剂不能提供更澄清的溶液。甲醇产生具有漂浮物的混浊悬浮液,而水产生均匀混浊的溶液。
