应用

该产品的典型用途是使贴壁细胞从培养表面脱落。使细胞从附着面脱落所需的胰蛋白酶浓度主要取决于细胞类型和培养阶段。
胰蛋白酶-EDTA溶液可用于以下应用:
  • 用作细胞培养物中的补充剂,以维持细胞生长
  • 在收获生长至融合状态的细胞
  • 时,用以使慢病毒转导的巨噬细胞脱落

生化/生理作用

胰蛋白酶可切割赖氨酸和精氨酸残基的C端肽。 如果酸性残基位于切割位点的两侧,则该反应的水解速度减慢,而如果脯氨酸残基位于切割位点的羧基侧,则不水解。 胰蛋白酶活性的最佳pH为7-9。 胰蛋白酶还可以切割氨基酸合成衍生物的酯键和酰胺键。 将EDTA作为螯合剂添加到胰蛋白酶溶液中,可中和掩盖胰蛋白酶作用肽键的钙镁离子。 去除这些离子可增加酶活性。

DFP、TLCK、APMSF、AEBSEF和抑肽酶等丝氨酸蛋白酶抑制剂可抑制胰蛋白酶。

组分

胰蛋白酶由含有223个氨基酸残基的单链多肽组成,由胰蛋白酶原在Lys - lle肽键处切去N-端六肽而得。 这些氨基酸序列通过6个二硫键交联。 这就是胰蛋白酶(即β-胰蛋白酶)的天然形式。 BETA-胰蛋白酶可自溶,在Lys-Ser残基裂解,产生α-胰蛋白酶。 胰蛋白酶是丝氨酸蛋白酶家族的成员。

注意

本品应置于-10至-40°C环境冷冻储存。避免反复冻融。

制备说明

用浓度过高的胰蛋白酶长时间孵育细胞会破坏细胞膜并杀死细胞。 溶解胰蛋白酶或由浓溶液稀释应使用不含Ca2+ 或Mg2+的缓冲盐溶液。
