Calcium/calmodulin-dependent protein kinase type IIa (UniProt: Q9UQM7; also known as EC:22.214.171.124, CaM kinase II subunit alpha, CaMK-II subunit alpha) is encoded by the CAMK2A (also known as CAMKA, KIAA0968) gene (Gene ID: 815) in human. CaM kinase II is one of the most widely studied multi-functional enzyme. It is found in high concentrations in the brain and in some regions, it may constitute up to 2% of the total protein content. CaM kinase II is a complex of about 12 subunits, which exist in four differentially expressed forms (a, b, g, d). The a and b isoforms are most abundant in neurons, whereas d and g isoforms are expressed in most tissues. CaM Kinase II is activated by Ca2+/calmodulin. Binding of calmodulin to calmodulin binding domain (aa 290-300) results in conformational change that relieves intrasteric autoinhibition and allows autophosphorylation of Thr-286 which turns the kinase in a constitutively active form and confers to the kinase a Ca2+-independent activity. CaM Kinase II is shown to be involved in synaptic plasticity, neurotransmitter release, long-term potentiation, and migration of developing neurons. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
