10% Neutral buffered formalin (NBF) is the most commonly used fixative throughout the world for light microscopy and is a somewhat forgiving fixative.[1] 10% NBF is well suited for large throughput laboratories, and requires a relatively short period of fixation, but can also be used for the long-term storage of tissue. Formalin fixation is thought to form between the aldehydes and the proteins, creating a gel, thus retaining cellular constituents in their in vivo relationship.[2] Once properly fixed, the tissue should be able to withstand the subsequent stages of tissue processing or staining.

Alkalmazás

Formalin solution, neutral buffered, 10% has been used as a popular general purpose histological fixative to fix tissues.[3][4][5]
Formalin solution, neutral buffered, 10% has been used in oil-red-o staining for the differentiation of 3T3-L1 preadipocyte cell line and mouse embryonic fibroblasts into adipocytes.[6] It has been used in the histological analysis of rabbit′s eye and diabetic rat intestine.[7][8]

    1. Neutral buffered formalin solutions are particularly sensitive to ambient light. Long term exposure to UV light may result in degradation. This product is packaged in natural opaque plastic. Note that solutions should be tightly closed during storage.

    1. Yes, this product contains Methanol which is included in the formaldehyde starting material. The concentration of methanol in this product is 1.0 - 1.5%.

  3. What are the differences between products 1004960700 and HT501128?

    1. Products 1004960700 (Formaldehyde solution, 4% buffered) and HT501128 (10% Neutral Buffered formalin) do not have a substantial difference. With HT501128, the formaldehyde used for dilution has a formaldehyde content of 37 to 40%. When the product is diluted 1 to 10 with water, it is referred to as 10% formalin, which has a formaldehyde content of 4% formaldehyde. Both products have the same concentration of formaldehyde in solution.

  4. Is there a 10% neutralized buffered formalin stating the osmolality?

    1. None of the formalin solutions are tested for osmolality. Osmolality is significant for live cells, as cells struggle to survive when their surroundings are vastly different from their own, on the other hand formalin is only a fixative solution. Formaldehyde solutions are buffered to prevent the formation of formalin pigment or acid hematin that forms when tissues are fixed at low pH.

  5. Is this product HT501128-4L is RNase/nuclease free?

    1. Currently, none of the formalin solutions are nuclease free. Products that are exo or endo nuclease free are typically labeled as suitable for "molecular" use. If no testing is shown as being performed, the products are not claimed to be nuclease free.

  6. Can the product HT501128-4L, Formalin solution, neutral buffered, 10%, be used for immunofluorescence staining?

    1. It is generally expected that if tissues are left in 10% Neutral Buffered Formalin for extended periods, the tissue may not be suitable for immunofluorescence. However, if the tissue spends only a short time in the 10% NBF, it might still be usable for immunofluorescence.
